Role of Exosomes Derived from Secretome Human Umbilical Vein Endothelial Cells (Exo-HUVEC) as Anti-Apoptotic, Anti-Oxidant, and Increasing Fibroblast Migration in Photoaging Skin Models

Background: Prolonged skin exposure to ultraviolet light rays leads photoaging, which is characterized molecularly by an increase in reactive oxygen species (ROS), cell apoptosis, and a decrease collagen. Photoaging therapy has been challenge until recently. Fibroblasts exposed B (UVB) proved be good model for photoaging skin. They are also the primary dermal cells that stimulate collagen production extracellular matrix (ECM), contribute aging. Exo-HUVEC rich growth factors, cytokines, miRNAs, they all play vital role cell-to-cell communication. The migration of fibroblasts crucial development, repair, regeneration tissue during repair aging.
 Objective: An vitro experimental study was conducted analyze effect on oxidative stress levels, fibroblast rate after UVB ray fibroblasts.
 Methods: cultures were divided into five groups, including one without exposure, with UVB+Exo-HUVEC at 0.1%, 0.5%, 1%, respectively. Oxidative levels measured using ELISA test malondialdehyde (MDA). Furthermore, flow cytometry used measure apoptosis PI/Annexin markers, while scratch assay examination imaging readings.
 Results: There significant differences MDA, PI/Annexin, between UVB-irradiated control group treatment (p<0.001).
 Conclusion: marker improvement, anti-apoptotic effects reduces stress, as well increases rate.
